I'm fairly sure that you aren't able to lower their defenses.
I can in no way prove this, but that has been the general consensus of the entire forums for months when this question comes up.
I'm specifically talking about ES.

ES has no def whatsoever.  so no, you can't lower it

2nd main skill but effect only enemy's first line. u cant reduce defence in es and ryoka cause they have 0.

What Mylar said is correct. While you "can" give them a lower defense debuff, it will do absolutely nothing because PvE targets such as ES do not have any defense value to begin with.

You can't on ryoka/ES. That's why we don't use 2nd skill and use 9th or 8th skill :)
