Muramasa is a support based Zanpakuto so no it won't help with Isshin
Any assaulter based or 'overall' zan will help out Isshin, so things like Senbon, Zangetsu, Tensa etc.
If you watch your battles you can see how much combo rate you get on your assaulters, so if it's quite low when it comes to your main, then use 8th skill, do NOT use 9th skill as it overwrites Senbon buff, which is higher than your main skill
if your main does a high enough combo rate for himself due to Isshin, then use your highest growth rate skill (either 2nd or 5th skill) ^^ :)

You seem like a smart fellow. So maybe I'll complicate the question. What about isshin and b.renji. Which would be better to cultivate?

muramasa will help Isshin some. Not as much as Zangetsu, but with that reiatsu growth, and the fact that he DOES give assaulters a decent bonus, it isn't ideal for him, but will do nicely. Tensa will boost him very nicely tho, lol
Use it all on main. You'll never replace him so make him useful.

Well they both have the same skill, I'd like to say SWP Aizen does indeed keep his new skill, or there is some sort of return points when you upgrade ^^ :)

If they are both in the same team?
B.Renji's skill is almost the same thing, just decrease enemy fury (And those he hits are pretty much going to die anyways, so no need for fury deduct)
It's quite hard to say which one i'd do if they was both in the same formation, but I'd mainly lean to Isshin to simply make more use of the soul cultivation skill, i'd rather make a Isshin that is somewhat useless become 'godly' than make an already 'god' (Renji ofc) well, apparently more of a god ^^
I am aware these skills upgrade, but still, this would mean going away from that insane amount of combo rate from Isshin, so making him still 'useless' due to no soul cultivation ^^ :)
