Depends if you can gain T1 on assulters only through kiske and rukia then they would be better. But if you can get T1 just from kiske on at least one partner than option one would be more beneficial, as yoruichis stun can be really nice in pvp.
Edit: And if no T1 or any plans for T1 in near future than again option 1.

I do have plans for T1 Rukia, only 1 fury stone to go, but well, T1 Rukia ain't gonna do much on her own will she...?

She can,  if she crits you'll only need to one or two fury stones on assulters for T1. And that can turn PVP around. Problem is making Rukia fast. Shes really fucking slow it hurts. Kiske is a bit faster but you need more fury for him. Personally I would run Rukia and kiske for PVE and Rukia (T1) and yoru for PVP.

if your close to a T1 Rukia, Stack her with your fury stones and your crit stones, T1 is a game changer in PVP, In fact, save up for Yama, a T1 stacked Yama will drop people 15 to 20 levels higher than you quite easily.  Make Rukia your fastest, the Yama (or another powerhouse assaulter) go as quick as you can as long as he's slower than rukia.  Tavern Kieske and Tavern Rukia are the only 2 with this bonus,  use it well, and win!!
