Because a partner can only combo once a round, no more, Isshin would be more game-breaker than Muken if we could combo infinitely.
Watched your void BR, there are two corrections to make your damage higher... One is the zanpakuto Ashizogi, [Filthy Frank's voice ON] THIS IS SHIT [OFF], the cute damage caused by the poison is nothing compared to Muken, get Tensa/Senbonzakura/Kyoka or any zanpakuto that raises the assaulters' damage, since the DMG buff multiplies the base DMG rather than adding, for example your Muken has 200% DMG, if he gets a buff from Isane his DMG becomes 340% (200 * 1.7) and not 270%, the more base DMG the partner has, the stronger is the boost from the buff, so equip your usual zanpakuto next time.
For your questions, ONLY the buff is a multiplier, the other sources of DMG (Halo, mod, hogyoku, talent, zanpakuto...) add into the base DMG. The Immunity Rate works the same.
And the other correction is : Make your main faster than Muken, we all know that if we attacks, the buff wears off (in your case the 50% ATK from Hiki) but the combo is magical, when a combo is triggered, you give an extra turn to the partner, and that extra turn is placed BEFORE the partner's original turn (where the buff wears off after) meaning that Muken attacks twice with the 50% ATK buff this way, this is important.
